Police and emergency services called to Richardson Road, Campvale, shortly after 9.30pm arrived at the scene to find two cars on fire after an accident.
Two backseat passengers in one of the cars died at the scene.
Four other occupants, two in each car, managed to escape and all were taken to John Hunter Hospital with serious injuries.
A crime scene has been established and an investigation into the cause of the crash is now underway.

Richardson Road was closed in both directions and was expected to be closed most of the night as investigators examined the accident.
In a busy night for a police, an accident occurred between a police car and another car during a pursuit at Alkira Avenue, Cessnock.
Police tried to pull over a car, with two children in it, after it allegedly ran a stop sign, just before 9pm.
The pursuit ended soon after in Alkira Avenue and there was an accident involving the vehicles but no one was injured. Two people were arrested at the scene.
Police said several other accidents yesterday afternoon were mostly single-vehicle incidents in which the driver appeared to have lost control.
A driver lost control on the Pacific Highway at Heatherbrae, near the Masonite Road intersection, and crashed into the median strip. No one was trapped or seriously injured.
Two other accidents occurred on the Pacific Highway, near Taree.
In Mulbring council workers rescued an Abernethy woman from the roof of her vehicle after she drove into floodwater just before 4.30pm.
A bridge at Horns Crossing Road at Vacy went under water because of the rain.
Across NSW the death toll was four for the long weekend.
